RETATUTRIDE Beyond the needle

The other night I sat asking AI how Retratutide worked. This is my Layman attempt to explain how the scientist engineer this molecule which I found facinating and thought I would share.
Retatrutide isn't made by gluing three different drugs together. It's one single long chain of 39 building blocks (amino acids) that was carefully redesigned so this one molecule can activate three different receptors in the body one at time but does all three (GLP-1, GIP, and glucagon).
Why make it one single chain instead of mixing separate pieces?
The three natural hormones are already "cousins" β they have very similar shapes. Scientists started with one of them and made small tweaks to the chain so it could fit into all three receptors.This gives the drug a consistent lifespan (about 6 days), so all three effects happen at the same time and you only need one weekly shot.
It lets them fine-tune the strength: the glucagon part is dialed down so it helps burn fat without spiking blood sugar (the GLP-1 and GIP parts keep things balanced).It's much simpler and safer to manufacture one molecule than trying to combine multiple separate ones.Why did they also have to "engineer it against DPP-4"?
Your body has a natural enzyme called DPP-4 that acts like scissors β it quickly cuts up natural GLP-1, GIP, and glucagon within minutes, making them useless.To stop this from happening to Retatrutide, the scientists made one tiny change: at position 2 on the chain at the N-terminus end they swapped in a special building block (called Aib) that acts like a shield. This blocks the DPP-4 enzyme from cutting the molecule.
They also added a fatty acid "tail" on Lys20 anchore (position 20) that helps the drug stick to a big protein in your blood (albumin). This hides it from being cleared out by the kidneys. The C20 Fatty acid act like a molecular "Velcro" that tighly binds to human serum albumin, the most abundant protein in the bloodstream thus escape the kidney and hides from other emzymes.

Together, these two changes let Retatrutide survive in the body for about 6 days instead of just minutes β which is why it can be injected just once a week.
In short:
One cleverly modified chain + a shield against the body's "scissors" enzyme + a fatty tail that hitches a ride throughout your body, delivering while evading = a long-lasting triple-action drug. -
It's absolutely amazing how these things work. You know this time last year, GLP1s and researching peptides was no where on my radar. Then started with a Telehealth company because my insurance doesn't cover glp1 meds for weight loss. Then I started going into deep dives, YouTube rabbit holes wanting to learn more about peptides.
