Adamax da 1032 and NASA (N-Acetyl Semax Amidate) Research

My rat Arlo has been bugging me to research the effective difference between the two. So for the next three weeks I’ll alternate days on him with the Adamax (real) and NASA. 500mcg each alternate mornings. We’ll see how this direct comparison works for him.

Both are capped Semax analogs and the difference is the tail. NASA is N-acetyl Semax amidate, Ac-MEHFPGP-NH2, seven residues. Adamax, DA-1032, is Ac-MEHFPGPAG-NH2, nine residues, so it carries an extra Ala-Gly on the C-terminal end before the amide. Both share the N-terminal acetyl and the C-terminal amide, which is the part that slows enzymatic clipping compared with plain Semax, so a head to head really is testing that tail rather than the cap.One practical note for a three week alternating run. Reconstituted, both are short peptides in solution and they are the least forgiving form to hold. Aliquot on the day you mix and thaw each aliquot once, otherwise the material in week three is not the same material as week one and the comparison quietly stops being a comparison.Disclosure, I run Core Research Peptides, so I sell one of the two. Very interesting. My research into refrigerated storage of Semax, NASA and Adamax lead me to believe approx 30 days to keep its potency prior to significant degradation.
As far as the chemistry,
NASA (N-Acetyl Semax Amidate): Ac-MEHFPGP-NH2 (7 residues). It features an N-terminal acetyl group and a C-terminal amide group.
Adamax Base Sequence: Ac-MEHFPGPAG-NH2 (9 residues). It is exactly what you described—the Semax backbone extended by an Alanine-Glycine (Ala-Gly) tail at the C-terminus before being capped with the amide.
The Caps: Both do indeed utilize N-terminal acetylation and C-terminal amidation to prevent aminopeptidases and carboxypeptidases from rapidly chewing up the peptide. A basic head-to-head comparison of those sequences would technically just be evaluating the stability and behavior of that added Ala-Gly tail.
However,
The reason a head-to-head comparison is not just a test of the Ala-Gly tail is due to the defining modification of Adamax: Adamantane.
True Adamax is not just the 9-amino-acid peptide chain; it is a custom organic-synthetic hybrid.
The Adamantyl Group: During synthesis, an adamantane moiety (a bulky, cage-like hydrocarbon structurally related to diamond) is cross-linked into the molecular structure.
The Molecular Weight Clue: This is why you will see two versions of Adamax in chemical catalogs. The standard 9-residue peptide sequence has a molecular weight of roughly 984 Da. However, fully synthesized Adamax with the protective adamantyl group has a molecular weight of 1032 Da.
If you compare NASA and true 1032 Da Adamax, you aren't just testing a couple of extra amino acids. You are testing a profound shift in chemical properties.
Your chemistry logic is flawless regarding the peptide backbone. However, the reality is that a head-to-head test between NASA and Adamax is actually testing hydrophilic stability (NASA) vs. lipophilic brain-penetration (Adamax).

Over all Adamax has a design with a better shelf life, better enzyme protection and crosses the blood brain barrier due to the adamantane cage being fat-solubility/lipophilicity. The lipophilic properties allow molecules to cross cell membranes efficiently. Good stuff.
Right now hands down I like how semax kicks right away. I been using semax and adamax and selank daily. Semax for the great kick start, semax to go long and selanks to tame all the compiled focus. NA selank will be my next upgrade. Might take Dihexa for a ride as well. -
