TL;DR — can one deep freeze liquid branded tirzepatide without loss of effectiveness or potency?

Long story — some researcher is sitting on a stash of branded tirzepatide pens that they got cheap due to mercy of insurance and employer. They switched to researching some other more effective molecules.

The branded T is in pens and have another year validity, but the researcher may not use all of it before that.

What if they take it out in a sterile vial and deep freeze it?

Where did you find that story @pep_researcher?

Standard warnings are to discard if frozen. I'd of course recommend not to do it. Either give away or toss it when the time comes. Just take the win if it was me, as tirz is cheap, not worth the risk.

Know it from someone I know 😜

They tried asking close friends who are more adventurous, but they too now do RUO research and don't want T.

The other friends are skeptical newbs who won't touch such molecules due to misconceptions like it will alter their brains, even if for the better. They don't want to lose their lovely food noise and binge eating. For the love of God! 😂

I've read instances on Reddit that people have frozen and used their compounded tirzepatide with no ill effects. Personally that's not my comfort level. But its people on Reddit. Take it for what it's worth.

I wouldn’t freeze it, assuming you mean indefinitely until you find some use for it or someone to buy it. Just use it or unload it before it expires. Not sure what the market for that looks like but it sounds sketchy.

@newenough @nuncles @researchcat Thanks. Yeah had huge doubts. But Redditters seemed super confident with their frozen compounded tirz, so a thought crept in... MAYBE!

Reg giving it away, yeah the person can't put it up on FB marketplace or eBay definitely. And within the closest trusted friends there are only two kinds — those fully submerged in RUO and new research, and others sticking to their food noise, binging, and conventional stuff... So no takers unfortunately.

@Stan-Douglas Nahh the person's not going to deal with random folks for this.

Also they are not sure if they want to get rid of the stock as yet. They started new research only recently, so undecided yet and so exploring ways to extend the molecule validity.

Honestly, I expect that it would be fine to use well past the expiration date. When I was buying compounded Tirzepatide, it regularly came with ‘extended BUD’ indicating it was good for 6 months or longer past its original best used by date. It won’t be good indefinitely, but it will be fine for a while. Unfortunately, it’s just a difficult item to offload.

My bride is finishing the last compounded vial I bought, which was bottled over a year ago and is past its extended BUD. It still works, though it may be less potent.

                      I love my tirz.

@ResearchCat Right. The expiration dates are usually very conservatively placed to avoid any liability issues.

The question was more about whether deep freezing the liquid form would extend expiration by several months/years, but doesn't seem to be the case.

Apparently there was some fringe study that found that doing freeze-thaw on GLP-3 made it lose much of the Glucagon component efficacy. GLP-1 seemed okay, but T also has GIP and there's no lab data on free-thaw on it.

Yeah, I would think 6 months after would be good still. Do these pens allow you to observe the med? Look for any cloudiness or changes? Never used one...

@Newenough No the auto injectors have less visibility. Unsure of the new KwikPens.

But no there's not so much worry about cloudiness and all if they are in sealed original containers.

I used to pick up my tirzepatide from the compounding pharmacy and it was always frozen. I asked why and was only told that they had to. It never felt any different from the name brand, though it was mixed with L-caritine

I'd be worried about water expansion in the pen probably more than the tirzepatide honestly.

Idk man I’d probably toss it. Is it still good? Maybe. I’d rather use a fresh vial.

I would love to see Randy or Peptide Crafters do a study on this. It would make sense for the peptide not to be affected, since lyophilization involves freezing, but freezing, as opposed to freeze drying, the crystallization could theoretically affect the peptide bonds.

It would be interesting to find out.

@kevinfay I have also received compounded tirz that was reconstituted with both saline and with bac water, which might make a difference. Both pharmacies said not to freeze and not to use if it did freeze. Might be nonsense. 🤷‍♂️

I have never frozen a 0.9% saline solution in lab but it could be that once the peptide is reconned, it would be subject to crenation once frozen and thawed due to osmotic pressure. Guessing...
